Western blot analysis of SLC7A1 (other name: Cat-1, and Slif in Drosophila) using SLC7A1 Rabbit Polyclonal Antibody (Product # PA5-42426) was performed by loading 10 mg of each cell lysates: HeLa (lane 1) and Drosophila S2 (lane 2) in sample buffer and 5 µL Spectra Multicolor Broad Range Protein Ladder (Product # 26634) onto a 10% SDS-PAGE gel. Proteins were transferred to PVDF membrane (Product # 88518) at 30V for 60 min on ice. Membrane was blocked in 5% Milk in PBST for 30 min at room temperature. The primary polyclonal SLC7A1 antibody (Product # PA5-42426) was used at a dilution of 1:500 in 5% Milk in PBST overnight at 4ºC. Followed by goat anti-rabbit alkaline phosphatase secondary antibody at a dilution of 1:20,000 for 1 hr. Chemiluminescent detection was performed using ECL substrate. Strong band was detected for HeLa cells (~ 67 kDa) and several bands for Drosophila (One corresponds with predicted 66 kDa band, second ~125 kDa could be a dimer).
